A haploid cdc2+ or cdc2-as colony was inoculated into YE medium at 25°C in the absence of analogue and the culture was sampled at various time points. Diploids were identified by their darker colour after replica-plating colonies to YE medium containing Phloxin B (Moreno et al., 1991). Samples were taken at the indicated cell densities, and after 1, 2 and 3 days in stationary phase (taken to be >2×107 cells/ml–1). At least 500 colonies were counted for each point
