Cover: A story of tails – a composite image showing different subcellular localizations of truncated versions of Jen1–GFP. Jen1 transporter tails are not simply unstructured domains, but play crucial roles in the many chapters of a transporter's life, including biogenesis, membrane trafficking, cell surface stability and transport activity. See article by C. Barata-Antuneset et al. (jcs260059).

RESEARCH HIGHLIGHTS
ESSAY
CELL SCIENCE AT A GLANCE
Chromosome organization through the cell cycle at a glance
Summary: The features of chromosome folding and organization are important for proper nuclear function. This Cell Science at a Glance article reviews chromosome and genome organization dynamics through the cell cycle.
REVIEW
Roles of the actin cytoskeleton in ciliogenesis
Summary: Overview of the recent studies defining the role of actin cytoskeleton during cilia formation in polarized epithelial cells.
SHORT REPORT
A cytoplasmic protein kinase couples engagement of Chlamydomonas ciliary receptors to cAMP-dependent cellular responses
Highlighted Article: A protein kinase in the cytoplasm is phosphorylated within 1 min after engagement of ciliary receptors during Chlamydomonas fertilization and is required to trigger the fusion-essential increase in cytoplasmic cAMP.
RESEARCH ARTICLES
Analysis of monocyte cell tractions in 2.5D reveals mesoscale mechanics of podosomes during substrate-indenting cell protrusion
Summary: Using a buckling model, we extend the current description of local podosome protrusion and include a mechanical explanation for protrusion on the cell scale.
Fluid shear stress regulates the survival of circulating tumor cells via nuclear expansion
Summary: This study reports that suspended tumor cells respond to fluid shear stress through histone acetylation-mediated nuclear expansion, which protects CTCs from shear-induced apoptosis.
Unveiling a novel serpinB2-tripeptidyl peptidase II signaling axis during senescence
Summary: A chemical probe-based activity assay was used to show that inhibition of tripeptidyl peptidase II (TPPII) by serpinB2 regulates proteostasis in senescent cells.
Interactions of cytosolic tails in the Jen1 carboxylate transporter are critical for trafficking and transport activity
Summary: Interaction of cytosolic terminal regions in the Jen1 carboxylate transporter, belonging to the major facilitator superfamily, are critical for trafficking and transport activity.
Vps68 cooperates with ESCRT-III in intraluminal vesicle formation
Summary: We show that yeast ESCRT-III interacts with Vps68, an endosomal membrane protein. We propose that Vps68 and ESCRT-III cooperate in the abscission of intraluminal vesicles by weakening the lipid bilayer.
The retromer complex regulates C. elegans development and mammalian ciliogenesis
Highlighted Article: Characterization of new roles for retromer in C. elegans development and in the regulation of ciliogenesis in mammalian cells, suggesting a novel role for retromer in CP110 removal from the mother centriole.
TOOLS AND RESOURCES
Single-cell analysis reveals the Comma-1D cell line as a unique model for mammary gland development and breast cancer
Summary: Characterization at a single-cell level of the unique heterogeneity of the mammary epithelial Comma-1D cell line, which illustrates its use as an experimental model to study both differentiation and transformation.
GIANI – open-source software for automated analysis of 3D microscopy images
Summary: General Image Analysis of Nuclei-based Images (GIANI) is a new plugin for the popular FIJI platform, designed for the automated analysis of 3D microscopy images of a wide range of sample types.

Fantastic proteins and where to find them – histones, in the nucleus and beyond
In this Review, Johanna Grinat and colleagues provide an evolutionary perspective of histones, nuclear chromatin and extracellular chromatin biology, describing the known extranuclear and extracellular functions of histones.

How to reduce your lab's carbon footprint
All stakeholders – from those working in the lab to those providing funding and infrastructure – have an important role to play to becoming more sustainable. In this Essay, Julie Welburn discusses what lab users can collectively do to transform biomedical research into a discipline that is significantly and positively sustainable.
