Cover: Mouse salivary gland acinar cells are an excellent model system for studying the dynamics of the actin cytoskeleton in vivo. The image shows a snapshot taken from intracellular intravital imaging of the surface of the salivary gland in an anaesthetised mouse expressing the tropomyosin isoform 3.1 tagged with mNeonGreen (green), and Lifeact–RFP to visualize actin (magenta). Myoepithelial cells are highly enriched in contractile actin cables while both tropomyosin and actin form an intricate cortical network at the basolateral plasma membrane of acinar cells. CELL SCIENCE AT A GLANCE
SUMO-specific proteases and isopeptidases of the SENP family at a glance
Summary: SUMO isopeptidases of the SENP family regulate the SUMO conjugation–deconjugation cycle and, hence, many cellular key functions. Here, we review the current knowledge on function and regulation of SENP proteins.
REVIEW
How cells sense their own shape – mechanisms to probe cell geometry and their implications in cellular organization and function
Summary: We discuss mechanisms on how cells read their own geometry, and the importance of these mechanisms in different physiological contexts.
SHORT REPORT
Parallel assembly of actin and tropomyosin, but not myosin II, during de novo actin filament formation in live mice
Summary: De novo assembly of functional actin–tropomyosin filaments in live rodents is investigated. Unlike non-muscle myosin II, tropomyosin is added to the actin filament shortly after nucleation.
RESEARCH ARTICLES
The feedback loop between miR-21, PDCD4 and AP-1 functions as a driving force for renal fibrogenesis
Summary: Anti-miRNA-21 oligonucleotides prevent progression of renal fibrosis by blocking the auto-regulatory loop between miR-21, PDCD4 and AP-1 during myofibroblast activation.
Puf3 participates in ribosomal biogenesis in malaria parasites
Summary: The novel nucleolar protein Puf3 in Plasmodium might have an essential function in biogenesis of the large ribosomal subunit rather than being involved in translational repression of mRNAs.
Rap1, Canoe and Mbt cooperate with Bazooka to promote zonula adherens assembly in the fly photoreceptor
Summary: Converging pathways regulate adherens junction stability and retention of Bazooka to promote zonula adherens maturation during epithelial cell morphogenesis in Drosophila.
Localization and functional consequences of a direct interaction between TRIOBP-1 and hERG proteins in the heart
Summary: TRIOBP-1 and the hERG ion channel protein directly interact and colocalize in cardiomyocytes. TRIOBP-1 overexpression affects channel distribution, IKr magnitude and the morphology of the action potential.
Ki-67 and condensins support the integrity of mitotic chromosomes through distinct mechanisms
Highlighted Article: Ki-67 and condensins, which localize to the external surface and the central axis of mitotic chromosomes, respectively, have independent yet cooperative functions in supporting the structural integrity of mitotic chromosomes.
The physiological regulation of macropinocytosis during Dictyostelium growth and development
Highlighted Article: A high-throughput flow cytometry assay is used to examine the regulation of macropinocytosis during transitions between feeding modes (macropinocytic and phagocytic) and entry into development in Dictyostelium. Macropinocytosis inhibitors are assessed.
Connexin hemichannels mediate glutathione transport and protect lens fiber cells from oxidative stress
Summary: Connexin hemichannels activated by oxidative stress in lens fiber cells mediate a cell protective mechanism through the uptake of reductants, maintaining lens transparency and homeostasis.
H2AFX and MDC1 promote maintenance of genomic integrity in male germ cells
Summary: H2afx and Mdc1 preserve male germ cell genome stability by promoting proper recombination-mediated synapsis, crossover formation and activation of the recombination-dependent checkpoint.
Hsp70–Hsp110 chaperones deliver ubiquitin-dependent and -independent substrates to the 26S proteasome for proteolysis in yeast
Summary: Hsp110 associates with the proteasome and ensures the efficient delivery of Hsp70 chaperone-associated proteins for proteolysis in the yeast S. cerevisiae.
Epidermal growth factor receptor and integrins control force-dependent vinculin recruitment to E-cadherin junctions
Summary: E-cadherin force transduction activates a kinase cascade via epidermal growth factor receptor and integrins that regulates force-dependent recruitment of VCL to stressed cadherin adhesions.
The protein kinase D1-mediated classical protein secretory pathway regulates the Ras oncogene-induced senescence response
Summary: The PKD1-mediated classical protein secretory pathway is involved in regulating senescence-associated secretory phenotype factor secretion and oncogene Ras-induced senescence.
Aplip1, the Drosophila homolog of JIP1, regulates myonuclear positioning and muscle stability
Summary: Aplip1 (the Drosophila JIP1) regulates both myonuclear position and muscle stability during early muscle development through different Dynein and Kinesin-regulated mechanisms.
